Effect of LPS on mRNA expression of inflammatory mediators and estradiol synthesis in granulosa cells from small/medium follicles. Bovine granulosa cells from small/medium diameter (2–8 mm) follicles were exposed to control medium (light bars) or LPS (1–104 ng/mL; dark bars) for 24 h. Gene expression of CXCL8 (A), IL6 (B), HSD17B (C) and CYP19A1 (D) were quantified by qPCR (n = 10), accumulation of 17β-estradiol (E) was quantified using ELISA (n = 10), and abundance of aromatase (F) was evaluated by western blot (n = 4). Gene expression data were log transformed for normality and a general linear model with fixed effect of LPS dose and random effect of replicate followed by least significant difference pairwise comparisons were used to analyze each dose compared to the medium alone control. Data are presented as the estimated marginal means ± S. E. M. * indicates P ≤ 0.05 compared to medium alone control.
